In this study, we assess the impact of chemical compounds on ovocyte maturation by preconditioning mouse ovarian surface culture (mOSC) in the presence of testosterone (T) and cyclic AMP (cAMP), which have long been used as simple methods to produce mature ovocytes (MOVCs) from OSCs (Párraga et al., 1995, 2000). We also evaluate whether the addition of a second combination (T + cAMP + fungizone Б(Fungizone)) is compatible with the inhibition of cAMP degradation by phosphodiesterase (PDE), which is responsible for cAMP catabolism. Mouse OSCs were pretreated with chemicals for 24 h and then transferred into TCM-199 medium supplemented with 10 ng/ml epidermal growth factor (EGF), 0.1 ng/ml follicle stimulating hormone (FSH), 1 microM steroidogenic hormones (T and 17 beta-estradiol (E2) and they were cultured further for 18 h. Trilostane (Tril) is an aromatase inhibitor (AI) that suppresses the conversion of T to E2. We found that the first combination (T + cAMP) and the Tril + cAMP combination both impaired the morphological maturation and up-regulation of the T receptor gene expression in MOVCs. On the other hand, addition of an inhibitory agent phosphodiesterase (PDE) in the culture system did not improve the production of viable MOVCs in combination with the second combination of hormones. Since the MOVCs cultured in the presence of T and FSH demonstrated maturation both morphologically and in terms of T receptor gene expression, we conclude that the biochemical changes in the live MOVCs are not the only important factors and that the addition of FSH to the culture system may influence the maturation of mouse ovocytes. The traditional method was shown to be completely inappropriate as a method to induce mouse ovocyte maturation in vitro, with the results suggesting that the addition of a second combination can improved the morphological and molecular aspects of the cultured cells, without the need for an inhibitory agent for cAMP degradation. d2c66b5586
